چکیده

Background: Whether AGR2 promotes adenocarcinoma growth as a secreted or ER localized protein is not known. Results: A unique C-terminal motif, KTEL, is required for AGR2 ER localization and function. Conclusions: Not all ER localization motifs are interchangeable. Significance: Specific ER localization signals may be required for protein function. AGR2's tumor promoting effects are mediated from the ER and not as a secreted protein.

AGR2 gene function requires a unique endoplasmic reticulum localization motif.

Soluble proteins are enriched in the endoplasmic reticulum (ER) by retrograde transport from the Golgi that is mediated by the KDEL receptors. In addition to the classic carboxyl-terminal KDEL motif, a variety of sequence variants are also capable of receptor binding that result in ER localization. Although different ER localization signals that exhibit varying affinities for the KDEL receptors...

Gating Behavior of Endoplasmic Reticulum Potassium Channels of Rat Hepatocytes in Diabetes

Background: Defects in endoplasmic reticulum homeostasis are common occurrences in different diseases, such as diabetes, in which the function of endoplasmic reticulum is disrupted. It is now well established that ion channels of endoplasmic reticulum membrane have a critical role in endoplasmic reticulum luminal homeostasis. Our previous studies showed the presence of an ATP-sensitive cationic...

A molecular specificity code for the three mammalian KDEL receptors

AC-terminal KDEL-like motif prevents secretion of soluble endoplasmic reticulum (ER)-resident proteins. This motif interacts with KDEL receptors localized in the intermediate compartment and Golgi apparatus. Such binding triggers retrieval back to the ER via a coat protein I-dependent pathway. To date, two human KDEL receptors have been reported. Here, we report the Golgi localization of a thir...

The Golgi-Localized Arabidopsis Endomembrane Protein12 Contains Both Endoplasmic Reticulum Export and Golgi Retention Signals at Its C Terminus C W

Endomembrane proteins (EMPs), belonging to the evolutionarily conserved transmembrane nine superfamily in yeast and mammalian cells, are characterized by the presence of a large lumenal N terminus, nine transmembrane domains, and a short cytoplasmic tail. The Arabidopsis thaliana genome contains 12 EMPmembers (EMP1 to EMP12), but little is known about their protein subcellular localization and ...

LAY, A New Endoplasmic Reticulum Retention Signal for UBIAD1 Protein

In this paper, we observed the co-localization of UBIAD1 and Ds Red-ER (an ER marker) in bladder carcinoma cell line T24 and human embryonic kidney cell line HEK293T. UBIAD1 accumulated on the endoplasmic reticulum (ER) in both T24 and HEK293T cell lines. Site-directed mutagenesis defined a LAY motif, positioned on 71-73 amino acids of the UBIAD1 N-terminus, as the ER retention signal of UBIAD1...
